Cleavage Under Targets and Tagmentation (CUT&Tag)

- Histone modification과 transcription factor의 genomic localization을 조사하기 위한 방법

 : 단백질과 DNA 사이의 상호작용 또는 관심 있는 단백질의 DNA 결합 부위를 확인

- 샘플의 양이 적거나 빠르게 실험을 진행해야 할 때 ChIP-Seq 대신 활용 가능

ChIP-Seq의 경우 고정된 chromatin을 sonicator 등으로 파쇄하고, IP를 진행하는데, CUT&Tag의 경우 고정되지 않은 세포를 concanavalin A bead에 붙이고, antibody incubation이 진행 된다. Antibody binding 이후에, chromatin은 잘려지고, NGS library는 sequencing adapter와 함께 준비된 protein A-Tn5(pA-Tn5) transposome enzyme을 사용한 tagmentation에 의해 준비된다. 

- 최소 5,000개의 세포로도 사용 가능

Not Require Fixation and Sonication

- Histone marks와 transcription factor 분석을 위해 개발되었으며, 최적화된 프로토콜 제공 

- Formaldehyde crosslinking으로 인한 artifacts가 발생하지 않음

Less Sequencing is Required for CUT&Tag 

- ChIP-Seq과 비교하여 시간과 비용을 절약하여 sequencing-ready libraries 제작이 가능

- Background signal이 낮아 sequencing depth를 줄일 수 있음

CUT&Tag-IT™ Service란?

- Low background signal의 장점을 가지고 있어 lower sequencing depth(2 million reads)를 사용하여 ChIP-Seq보다 적은 비용으로 고품질 데이터 생성 가능

- End-to-end CUT&Tag-IT™ Service 이용 시, 프로토콜을 최적화할 수 있고, 여러 항체를 테스트하여 효과가 있는 항체를 찾을 수 있으며, next-generation sequencing data와 결과에 대한 설명이 기본적으로 포함되어 있어 별도의 생물정보학 학습이 필요하지 않음
