기존의 복잡하고 오랜 시간을 요구하는 IHC, FISH 검사법을 대체할 수 있음

(해외에서는 간단한 진단검사법으로 ddPCR을 제안하는 논문이 발표 되고 있는 추세임)

* Droplet Digital^TM PCR이란?

Droplet Digital^TM PCR System은 20ul의 PCR 반응을 2만개 droplet으로 쪼개어 증폭시킨 후, target DNA를 계수하는 시스템이다.

Droplet에서의 target DNA의 증폭 여부에 따라 positive droplet(1)과 negative droplet (0)으로 digital signal처럼 받아들여 계수하고,

프아송 분포를 통해 target DNA의 copy를 계산해 최종적으로 샘플 ul당 Copy 수로 결과 값을 확인할 수 있다.

QX200^TM Droplet Digital^TM PCR Workflow

- 분석하고자 하는 시료와 probe(FAM, HEX/VIC) 또는 EvaGreen 형광염료를 포함하는 PCR 반응액을 20 ul로 준비한다.

- 준비한 PCR 반응액과 오일을 카드리지에 분주하여 Droplet Generator에 장착하면 2분 만에 자동으로 각 샘플 당 2만의 droplets이 생성된다.

- 하나의 카드리지는 최대 8개 시료까지 사용할 수 있다.

- 생성된 droplets을 96-well PCR plate로 옮긴 후 conventional PCR을 통해 PCR 반응을 진행한다. PCR 반응이 완료되면 Plate를 Droplet Reader로 장착한다.

- Reader와 연동된 QuantaSoft를 통해 최대 96개 샘플의 형광 신호를 각각 감지를 통해 positive와 negative droplet 및 타겟 유전자의 copy 수를 계산하여 분석한다.
